Akonni has developed a portable automated DNA/RNA purification platform, the TruTip Automated Workstation, which can homogenize difficult samples such as sputum, stool and tissue, followed by concentration and purification of nucleic acid. The unit comes in an enclosed cabinet with HEPA filtration for Class 1 bench top use, and is small enough to fit within a Class 2 biosafety laminar flow hood for secondary containment. The workstation runs up to 8 samples in parallel. Purified nucleic acids from the workstation have been demonstrated to be compatible with downstream diagnostic approaches such as qPCR, dPCR, electrophoresis, isothermal amplification methods (i.e. LAMP, HDA, TDA), line probe assays, microarrays, and sequencing.
The main design features include 1) the MagVor™ homogenization step for difficult to liquefy samples, 2) a heating block for traditional chemical and or heating lysis and decontamination, and 3) the TruTip technology for nucleic acid concentration and purification from small to large volume samples, all at a highly competitive instrument price and consumable cost.
The TruTip Automated Workstation includes a patented non-contact bead beating sample homogenization technology. MagVor (i.e. Magnetically-induced Vortexing) consists of a rapidly rotating magnet and a disposable tube, which includes a disposable magnetic stir disk and glass beads. Rotation of the magnet induces an alternating magnetic field in the tube, causing the magnetic stir disk to spin and agitate the glass beads. At high rotational velocities, the glass beads undergo chaotic mixing, which results in specimen homogenization and cell disruption. The absence of physical contact between the magnet on the instrument and internal magnetic disk in the tube simplifies the mechanical interface of this approach, and eliminates heat transfer (in contrast to a sonication horn). The unique design also eliminates decoupling of the external and internal magnets.
